Semaglutide vs Tirzepatide vs Teriparatide vs Abaloparatide vs Glycopyrrolate: Peptide API Comparison — CAS, Synthesis Route, Patent Cliff & Export Volume
One of these five weighs 398 daltons and is finished in two synthetic steps. The other four weigh between 3,960 and 4,814 daltons and are assembled one amino acid at a time. Glycopyrrolate (CAS 596-51-0) is a quaternary ammonium anticholinergic made by classical organic synthesis, shipped at room temperature and free of any compound patent since the 1960s. Teriparatide (CAS 52232-67-4) and abaloparatide (CAS 247062-33-5) are 34-residue bone-anabolic peptides dosed in micrograms. Semaglutide (CAS 910463-68-2) and tirzepatide (CAS 2023788-19-2) are lipidated incretin peptides dosed in milligrams, and their demand is the reason China's peptide reactor capacity moved from hundred-kilogram to tonne scale in three years. The five share a category label — injectable prescription API — and almost nothing else.
At a Glance
Family: one synthetic small molecule and four synthetic peptides, spanning three receptor classes — muscarinic, PTH1R and incretin.
Common mechanism of the four peptides: none. Each engages a different G-protein-coupled receptor, and the two bone peptides act on the same receptor through different conformational states.

Three groupings matter more than any individual row. The anticholinergic group is glycopyrrolate alone. The PTH1R group is teriparatide and abaloparatide. The incretin group is semaglutide and tirzepatide. Capacity, cold chain, documentation depth and capital intensity all follow the group, not the individual molecule.
2. Two Manufacturing Universes: Flask Synthesis vs Resin Synthesis
The dividing line in this comparison is not therapeutic category. It is whether the molecule is built in solution or on a solid support.
Glycopyrrolate: classical ester synthesis
- Route. Esterification of a cyclopentylmandelic acid derivative with a 3-hydroxy pyrrolidine, followed by quaternisation with methyl bromide to give the permanently charged pyrrolidinium. Two bond-forming operations, both established for decades.
- Purification. Crystallisation. No preparative chromatography at commercial scale, no lyophilisation.
- Physical form and handling. White crystalline powder, melting point 191–195 °C, freely soluble in water, methanol and ethanol, practically insoluble in chloroform and ether, log P −1.18. Ambient storage, no cold chain.
- Scale economics. Gram-per-patient dosing means kilograms to tonnes of API per year globally, produced by a broad multi-source base at generic prices.
The four peptides: stepwise assembly and chromatographic purification
- Route. Fmoc solid-phase peptide synthesis (SPPS), liquid-phase or hybrid fragment condensation, or recombinant expression. Each residue is coupled and deprotected in a cycle; for a 39-residue chain such as tirzepatide, that is 38 couplings before cleavage.
- Yield arithmetic. Coupling yield compounds across the chain. A 99.5% per-step yield over 38 couplings returns roughly 83% before any purification losses, which is why crude purity, and therefore purification load, is the dominant cost driver for the long peptides.
- Post-chain chemistry. Semaglutide and tirzepatide additionally carry a lipidated side chain installed through a linker on a lysine ε-amino group. The lipidation step and the linker chemistry are separate unit operations on top of chain assembly.
- Purification and isolation. Preparative reversed-phase HPLC followed by lyophilisation. Purification, not synthesis, is usually the capacity bottleneck, and it is expressed in reactor volume and column-hours rather than in flask size.
Impurity universes
The two technologies generate structurally different impurity profiles, and an analytical package built for one is not transferable to the other.
- Small molecule. Process-related impurities, residual starting materials and reagents, diastereomeric ratio, residual solvents under ICH Q3C, elemental impurities under ICH Q3D, polymorph and particle-size control.
- Peptide. Deletion and insertion sequences, incompletely deprotected chains, D-isomers from racemisation during coupling, oxidised methionine, aspartate isomerisation and deamidation, truncated and acetylated species, aggregates, plus counter-ion (acetate or trifluoroacetate) and residual water.

Identity traps in this group
- Glycopyrrolate exists as two different masses with one CAS number. The bromide salt is C19H28BrNO3 at 398.33 g/mol; the glycopyrronium cation is 318.43 g/mol. The FDA label states that the active moiety, glycopyrronium, constitutes 80% of the nominal tablet strength — 1 mg of glycopyrronium bromide corresponds to 0.8 mg of glycopyrronium. A specification written against one basis and a lot assayed against the other produces an 20% discrepancy.
- USAN and INN differ for the same substance. Glycopyrrolate is the United States Adopted Name; glycopyrronium is the International Nonproprietary Name. Both refer to CAS 596-51-0 in its bromide form.
- Teriparatide and abaloparatide are both 34 residues and both act at PTH1R, and are not substitutes. Sequence homology between the two is 41%. They favour different receptor conformations and different signalling durations, and they differ in labelled dose, presentation and patent position.
- Abaloparatide is listed with two hydrogen counts in circulation. The FDA label gives C174H300N56O49 at 3961 daltons; several chemical databases carry C174H299N56O49 at approximately 3959.6. Both resolve to CAS 247062-33-5, and the difference changes the fourth significant figure of the mass.
- Semaglutide and tirzepatide are both lipidated, and the lipid chains differ. Semaglutide carries a C18 fatty diacid through an AEEAc-AEEAc-γ-Glu linker; tirzepatide carries a C20 fatty diacid directly on a lysine side chain. Neither is a generic version of the other, and neither shares an impurity profile with an unlipidated peptide.
- Semaglutide's molecular weight is published across a narrow range. Values from 4113.58 to 4113.64 g/mol appear across reference sources, depending on whether average or monoisotopic mass is reported. The mass basis belongs in the analytical method description.
4. Glycopyrrolate — The 398-Dalton Quaternary Ammonium
Glycopyrrolate is the control case in this comparison: the only molecule that is made by ordinary synthetic chemistry, stored at room temperature and priced as a mature generic.
Chemistry and pharmacology
- Structure. Pyrrolidinium, 3-[(cyclopentylhydroxyphenylacetyl)oxy]-1,1-dimethyl-, bromide. The permanently charged quaternary ammonium is the reason the molecule does not cross the blood–brain barrier to a significant degree, which is the pharmacological basis for its comparatively low central anticholinergic burden relative to atropine or scopolamine.
- Stereochemistry. The substance contains two chiral centres and is supplied as a racemate of a 1:1 mixture of the R,S and S,R (threo) diastereomers. Both diastereomers are pharmacologically active.
- Physicochemical profile. White crystalline powder, melting point 191–195 °C, freely soluble in water, methanol and ethanol, practically insoluble in chloroform and ether, log P −1.18.
Labelled presentations and where the API goes
| Route / presentation | Labelled context | API demand character |
|---|---|---|
| Oral tablets (1 mg, 2 mg) and orally disintegrating tablets | Adjunct in peptic ulcer disease; a label dating to 1961 and largely superseded in practice by acid-suppressing agents | Mature, price-driven, multi-source |
| Injectable solution (0.2 mg/mL) | Reduction of salivary, tracheobronchial and pharyngeal secretions; blockade of cardiac inhibitory reflexes during induction and intubation; protection against the peripheral muscarinic effects of neostigmine and pyridostigmine given to reverse neuromuscular blockade | Hospital channel, subject to sterile-manufacturing capacity constraints |
| Oral solution (1 mg/5 mL, Cuvposa) | Chronic severe drooling in patients aged 3–16 years with neurologic conditions | Narrow, formulation-dependent |
| Topical cloth (2.4%, Qbrexza, approved June 2018) | Primary axillary hyperhidrosis in adults and children aged 9 and older | Small-mass, premium-priced presentation |
| Inhalation powder (15.6 mcg, Seebri Neohaler), nebuliser solution (25 mcg/mL, Lonhala Magnair) | Long-term maintenance treatment of airflow obstruction in COPD | Device-bound; per-unit API mass is very low |
| Fixed combinations (with formoterol, with indacaterol, budesonide/glycopyrrolate/formoterol) | COPD maintenance; treatment migration is toward dual and triple therapy | Highest-value channel, protected by device, evidence and regulatory package rather than by the molecule |
5. Teriparatide — PTH(1-34), the Bone-Anabolic Reference
Teriparatide is the 34 N-terminal amino acids of human parathyroid hormone. It is the reference compound for the entire bone-anabolic category and the molecule with the longest post-patent history in this comparison.
| Parameter | Teriparatide |
|---|---|
| CAS / formula / MW | 52232-67-4 / C181H291N55O51S2 / 4117.72 g/mol (free base) |
| ATC code | H05AA02 |
| Labelled regimen | 20 µg subcutaneous once daily |
| Bioavailability / half-life | ≈95% subcutaneous; ≈1 hour subcutaneous |
| Drug product presentation | Pre-filled pen, 2.4 mL cartridge, 28 doses of 20 µg in 80 µL; pH 4 acetate buffer with metacresol as preservative |
| Storage of the finished product | 2 °C to 8 °C, do not freeze; in-use chemical, physical and microbiological stability demonstrated for 28 days; the pen is capped after use because the solution is light-sensitive |
| Stability-relevant sequence feature | Two methionine residues; methionine oxidation reduces activity, and methionine-containing peptides are handled under oxygen-limited conditions |
| Originator commercial context | Forteo reported worldwide sales of USD 1.6 billion in 2018, before follow-on entry |
Post-patent history
- Patents on the originator product expired in both Europe and the United States in August 2019.
- United States. PF708 was approved by the FDA on 4 October 2019 through a 505(b)(2) new drug application referencing Forteo, on the basis of a 24-week study in 181 patients with osteoporosis; it was launched as Bonsity in June 2020. Teriparatide was not on the FDA's transition list to biologic regulation, which is why the follow-on route was 505(b)(2) rather than a biosimilar application, and why a therapeutic-equivalence rating rather than an interchangeability designation was the available outcome.
- Europe. Biosimilar teriparatides from Stada Arzneimittel and Gedeon Richter were authorised in January 2017; Qutavina and Livogiva in August 2020; Sondelbay in March 2022; Kauliv in February 2023.
- Canada. Osnuvo was approved in January 2020.
6. Abaloparatide — PTHrP(1-34) Analog and the RG Conformation
Abaloparatide and teriparatide are the same length, act at the same receptor and are the two molecules most often mistaken for one another in enquiries. The difference is conformational, and it is the reason both exist.
| Parameter | Abaloparatide |
|---|---|
| CAS / MW | 247062-33-5 / 3961 daltons (FDA label, C174H300N56O49) |
| Origin | Synthetic analog of human parathyroid hormone-related peptide, PTHrP(1-34) |
| Homology | 41% to hPTH(1-34); 76% to hPTHrP(1-34) |
| Receptor pharmacology | PTH1R agonist; preferentially engages the RG (G-protein-coupled) conformation, producing a shorter, more transient cAMP response |
| Contrast with teriparatide | Teriparatide binds the R0 conformation with relatively high affinity, so the ligand–receptor complex is internalised and continues to generate cAMP from endosomes |
| Labelled regimen | 80 µg subcutaneous once daily |
| Presentation | Disposable single-patient-use pen; cartridge contains 1.56 mL with 3.12 mg abaloparatide; 30 daily doses of 80 µg in 40 µL |
| Formulation | Acetate buffer at target pH 5.1–5.2 with phenol as preservative |
| Approval | United States, 2017 |
Non-coded residues are a manufacturing note for both bone peptides: abaloparatide contains Aib (2-aminoisobutyric acid), a sterically hindered residue that complicates coupling efficiency at that position, and carries a C-terminal amide rather than a free acid.
7. Semaglutide — The Lipidated GLP-1 That Rescaled Peptide Capacity
Semaglutide is the molecule that moved peptide API from a specialty category into a capacity-planning problem for the entire Chinese API sector.
| Parameter | Semaglutide |
|---|---|
| CAS / formula / MW | 910463-68-2 / C187H291N45O59 / 4113.58 g/mol |
| Chain | 31 amino acids; 94% sequence homology to native GLP-1 |
| Engineering | Aib at position 8 confers resistance to DPP-4; acylation of Lys26 with a C18 fatty diacid through an AEEAc-AEEAc-γ-Glu linker confers albumin binding |
| Resulting pharmacokinetics | Half-life of approximately one week; plasma protein binding above 99%; extensive metabolism before excretion in urine and faeces |
| Approvals | Ozempic (injectable, type 2 diabetes) 5 December 2017; Rybelsus (oral, type 2 diabetes) 20 September 2019; Wegovy (injectable, chronic weight management) 4 June 2021; Wegovy cardiovascular risk reduction 8 March 2024; chronic kidney disease in type 2 diabetes 30 July 2025 |
| Oral presentation | Rybelsus combines semaglutide with the absorption enhancer sodium N-(8-[2-hydroxybenzoyl]amino)caprylate (SNAC), which raises trans gastric epithelial permeability and provides local pH modulation that protects the peptide from gastric proteolysis |
| US customs classification | CBP ruling NY N341149 (2024): bulk semaglutide classified in HTSUS 2937.19.0000, duty free |
Capacity consequences
- Purification, not synthesis, sets the ceiling. The lipidation step and the linker add unit operations after chain assembly, and preparative HPLC throughput is the binding constraint for every manufacturer in the category.
- Capacity is being built in reactor litres. Published figures for the Chinese sector show SPPS reactor volume rising from 41,000 L at the end of 2024 toward a stated 100,000 L target at one CDMO, and from roughly 30,000 L toward 44,000 L at another, alongside tonne-scale workshops with single-batch outputs above 10 kg for long-chain modified peptides such as semaglutide and tirzepatide.
- Alternative routes are being industrialised. Fermentation-based peptide production has been reported at pilot scale for semaglutide API, with stated plans well above current output; if the cost basis works at scale, it changes the purification economics rather than the molecule.
8. Tirzepatide — Dual GIP/GLP-1 and the Longest Patent Runway
Tirzepatide is the longest chain in this comparison, the largest molecule by mass, and the one with the longest remaining period of patent protection.
| Parameter | Tirzepatide |
|---|---|
| CAS / formula / MW | 2023788-19-2 / C225H348N48O68 / 4813.5 g/mol |
| Development code | LY3298176 |
| Chain | 39 amino acids; Aib at positions 2 and 13; C-terminal amide |
| Engineering | C20 fatty diacid side chain on a lysine residue for albumin binding; dual activity at GIP and GLP-1 receptors |
| Approvals | Mounjaro (type 2 diabetes) 13 May 2022; Zepbound (chronic weight management) 8 November 2023; Zepbound for moderate-to-severe obstructive sleep apnea in adults with obesity 20 December 2024 |
| Commercial scale | Combined Mounjaro and Zepbound revenue exceeded USD 10.1 billion in the third quarter of 2025, up 132% year on year, passing the previous highest-selling pharmaceutical product |
| US customs classification | CBP ruling N341897 (16 August 2024): bulk powder tirzepatide classified in HTSUS 2937.19.0000, duty free |
| Patent position | Compound patents run to 2036 in both China and the United States, with peripheral patents extending to 2039–2041 |
9. Dose Mass: Why One API Needs Tonnes and Another Needs Kilograms ★
Labelled dose multiplied by dosing frequency gives a quantity that no market report states and that every capacity plan depends on: the mass of API consumed by one treated patient in one year. The figures below are arithmetic from published label regimens and are stated before titration waste, overfill, rejection and process yield.
| Compound | Regimen used for the calculation | API per treated patient-year | Order of magnitude | Capacity implication |
|---|---|---|---|---|
| Glycopyrrolate | 2 mg orally three times daily | ≈ 2,190 mg (2.19 g) | Grams | Kilogram-to-tonne campaigns; classic generic scale |
| Tirzepatide | 15 mg subcutaneous once weekly (maximum maintenance dose) | ≈ 780 mg | Hundreds of milligrams | Populations of millions require multi-tonne purified capacity |
| Semaglutide | 2.4 mg subcutaneous once weekly (Wegovy maintenance) | ≈ 125 mg | Hundreds of milligrams | Same order as tirzepatide at higher patient numbers |
| Abaloparatide | 80 µg subcutaneous once daily | ≈ 29.2 mg | Tens of milligrams | Kilogram-scale annual requirement |
| Teriparatide | 20 µg subcutaneous once daily | ≈ 7.3 mg | Single-digit milligrams | The smallest API mass per patient in the group |
10. Global Export Volume Ranking (China API Supply) ★
The decisive question in this category is which of the five is actually moving cross-border volume. The ranking below is a qualitative index based on relative API trade activity — weighted by transaction value and enquiry density rather than by shipped mass — from Chinese and other Asian supply chains. It is a directional market view, not a published statistical series, and the ordering separates the two incretins from the remaining three by more than an order of magnitude.
- Semaglutide (CAS 910463-68-2) — the largest peptide API line in the group by a wide margin. Diabetes, weight management, cardiovascular risk reduction and chronic kidney disease indications stack on the same molecule, and the China compound patent expired on 20 March 2026 while protection in the United States, Europe and Japan runs to 2031–2032. More than ten domestic Chinese filers had submitted generic or biosimilar applications, with further filings reported from Qilu, CSPC, Chengdu Beite, Huadong Medicine and United Laboratories, and chemically-modified generics expected from the third quarter of 2026. Demand spans GMP API, clinical-trial material and research-grade channels, and the qualified-supplier field is still narrow relative to the enquiry volume.
- Tirzepatide (CAS 2023788-19-2) — comparable demand, longer runway, tighter supply. Combined originator revenue exceeded USD 10.1 billion in a single quarter of 2025. Compound protection runs to 2036 in China and the United States with peripheral patents to 2039–2041, so there is no lawful generic channel in any major market, and API movement is concentrated in clinical, contract-manufacturing and research channels. At 39 residues it is also the most synthesis-intensive molecule in the group.
- Glycopyrrolate (CAS 596-51-0) — the only high-mass, low-unit-value line. No patent, no cold chain, mature multi-source competition and steady hospital, respiratory and dermatology demand. By shipped mass it is plausibly the largest of the five; by value per kilogram it is the smallest by two to three orders of magnitude. Its position in the index reflects transaction value and enquiry density rather than tonnage.
- Teriparatide (CAS 52232-67-4) — mature, post-patent, kilogram-scale. Originator patents expired in 2019 and follow-on or biosimilar products are authorised in the United States, the European Union and Canada. Volume is constrained by the label rather than by supply: at 20 µg daily, annual API requirement is measured in single-digit kilograms for a patient population in the hundreds of thousands. Competition among API suppliers is on documentation status and on the impurity package rather than on capacity.
- Abaloparatide (CAS 247062-33-5) — the smallest line in the group. A 2017 approval with a narrower labelled population and a smaller installed base than teriparatide, and the lowest enquiry density of the five. Realistic orders are kilogram-scale or below, and the manufacturing barrier — Aib incorporation, C-terminal amidation, 34-residue purification — is high relative to the volume, which keeps unit cost elevated.
11. Patent Position & the March 2026 Semaglutide Cliff in China ★
Patent status, not chemistry, determines which of these molecules can be lawfully sourced as a generic API and in which market. The spread across the five is the widest single differentiator in the group.
| Compound | Patent position | Consequence for API sourcing |
|---|---|---|
| Semaglutide | Core compound patent expired in China on 20 March 2026. Protection in the United States, Japan and Europe runs to 2031–2032; formulation patents in some jurisdictions extend as far as 2042 | Two-speed market. Generic and biosimilar filings are live in China, India and Brazil; the United States remains closed until December 2031. The originator has announced price reductions of 35–50% across its line for 2027, which compresses the price headroom available to entrants |
| Tirzepatide | Compound patents to 2036 in both China and the United States; peripheral patents to 2039–2041 | No meaningful generic channel before the mid-2030s in any major jurisdiction. API movement is confined to the originator's supply chain, contract manufacturing, clinical supply and research channels |
| Teriparatide | Originator product patents expired in August 2019 in both the United States and Europe | Fully post-patent. Follow-on products approved from 2019 in the United States and from 2017 in the European Union, with biosimilars authorised through 2023 |
| Abaloparatide | Later approval (2017) and therefore a later exclusivity runway than teriparatide | Generic API demand is limited and confined to markets where the relevant protection has lapsed |
| Glycopyrrolate | No compound patent. Protection, where it exists, attaches to formulation, combination and device | Free to source as an API. Competition is on dosage form, device and regulatory package rather than on molecule rights |
A second, separate regulatory boundary: compounding channels
Shortage-driven compounding was a material demand channel for both incretins in the United States, and that channel has closed. The FDA declared semaglutide injection in shortage in August 2022 and tirzepatide injection in December 2022; both were removed from the shortage list in late 2024 — tirzepatide in October and semaglutide injection in December. Enforcement discretion for compounding then ended on a staggered schedule: 503A pharmacies for tirzepatide in February 2025, 503B outsourcing facilities for tirzepatide in March 2025, 503A pharmacies for semaglutide in April 2025 and 503B facilities for semaglutide in May 2025.
12. Solubility, Delivery & Formulation Reality
Each of the five presents a different delivery problem, and in four of the five the delivery problem is the reason the product exists in its current form.
| Compound | Aqueous behaviour / presentation | Delivery | What the developer inherits from the API specification |
|---|---|---|---|
| Glycopyrrolate | Freely soluble in water, methanol and ethanol; practically insoluble in chloroform and ether | Oral, intravenous, intramuscular, topical and inhaled, plus fixed-dose inhaler combinations | Diastereomeric ratio, crystal form, particle-size distribution for inhaled grades, residual solvents |
| Teriparatide | Supplied as a preserved solution at pH 4 in acetate buffer with metacresol | Subcutaneous, once daily, pre-filled pen | Methionine-oxidation level, aggregate and particulate control, light-protection requirements, counter-ion and peptide content |
| Abaloparatide | Supplied as a preserved solution at pH 5.1–5.2 in acetate buffer with phenol | Subcutaneous, once daily, disposable pen | Aib-incorporation completeness, amidation completeness, aggregate control, pH-range conformance |
| Semaglutide | Injectable solution in pre-filled pen; oral tablet co-formulated with the absorption enhancer SNAC | Subcutaneous weekly; oral daily in the SNAC presentation | Lipidation completeness and linker integrity, albumin-binding-relevant purity, oxidation and deamidation profile |
| Tirzepatide | Injectable solution in pre-filled pen or vial | Subcutaneous weekly | Lipidation completeness, deletion-sequence profile from 38 couplings, aggregate control |
13. Synthesis Difficulty, Storage & Cold Chain

Degradation pathways worth specifying
- Oxidation. Teriparatide carries two methionine residues, and methionine oxidation reduces activity; methionine-containing peptides are handled under oxygen-limited conditions.
- Freeze–thaw. Repeated freeze–thaw of peptide solutions inactivates a fraction of the material each cycle; aliquoting after first dissolution is the standard laboratory handling response. The approved teriparatide solution must not be frozen and must not be used if it has been frozen.
- Aggregation and precipitation. Reconstituted lyophilised PTH(1-34) has been reported to precipitate within two to four weeks depending on concentration and temperature. Peptide concentration and temperature history belong in the stability protocol.
- Light. Light protection is written into the handling of teriparatide at the finished-product level, and light-protected storage is standard for the lyophilised peptides generally.
- Counter-ion. Acetate and trifluoroacetate are the two counter-ions encountered after preparative HPLC. The identity and quantity of the counter-ion affect peptide content, solubility and the residual-solvent statement.

15. API Selection Guide for Formulation Projects
- Which manufacturing universe does the project sit in? Small-molecule synthesis with crystallisation and ambient storage, or solid-phase assembly with preparative chromatography and lyophilisation. Capital, lead time and analytical depth diverge at this point and everything downstream follows.
- Is the target a peptide at all? Glycopyrrolate is the only non-peptide in the group, and it is the only one with no cold chain, no compound patent and a broad multi-source base.
- Is the receptor PTH1R or an incretin receptor? Teriparatide and abaloparatide are bone-anabolic PTH1R agonists with microgram dosing and kilogram-scale annual API requirement. Semaglutide and tirzepatide are incretin-receptor agonists with milligram dosing and tonne-scale requirement.
- Does the receptor-conformation profile matter to the programme? Teriparatide engages R0 relatively strongly, giving prolonged cAMP signalling; abaloparatide is RG-selective, giving a shorter response. The two are the same length and the same receptor and are not interchangeable.
- Which market is the finished product destined for? Semaglutide's compound patent lapsed in China in March 2026 and runs to December 2031 in the United States; tirzepatide runs to 2036 in both. The lawful sourcing position is set by the destination, not by the molecule.
- What is the declared end use? Licensed finished-dose manufacture with an open DMF or CEP, clinical supply, or research use occupy different regulatory positions, and United States compounding enforcement discretion for both incretins ended during 2025.
- Is the mass basis defined? Glycopyrrolate has a bromide-salt and a cation basis differing by 20%; each peptide carries a counter-ion and a variable water content. Purity, peptide content, water and counter-ion belong in the specification together.
- Incretin injectable programme, China or India or Brazil destination? → Semaglutide, CAS 910463-68-2.

- Bone-anabolic programme with RG-selectivity rationale? → Abaloparatide, CAS 247062-33-5.
- Anticholinergic programme across oral, injectable, topical or inhaled routes? → Glycopyrrolate, CAS 596-51-0.
16. Frequently Asked Questions
What is the difference between semaglutide and tirzepatide at the API level?
Semaglutide (CAS 910463-68-2, C187H291N45O59, 4113.58 g/mol) is a 31-residue GLP-1 receptor agonist with Aib at position 8 and a C18 fatty diacid attached to Lys26 through an AEEAc-AEEAc-γ-Glu linker. Tirzepatide (CAS 2023788-19-2, C225H348N48O68, 4813.5 g/mol) is a 39-residue dual GIP and GLP-1 receptor agonist with Aib at positions 2 and 13, a C20 fatty diacid side chain and a C-terminal amide. They differ in chain length by eight residues, in receptor coverage, in lipid chain and linker chemistry, and in patent position: semaglutide's China compound patent expired in March 2026, while tirzepatide's runs to 2036.
Are teriparatide and abaloparatide interchangeable?
No. Both are 34-residue PTH1 receptor agonists, but sequence homology between them is 41% — abaloparatide is a PTHrP(1-34) analog with 76% homology to hPTHrP(1-34), while teriparatide is identical to human PTH(1-34). They also differ pharmacologically: teriparatide binds the R0 receptor conformation with relatively high affinity, producing prolonged cAMP signalling from internalised complexes, whereas abaloparatide preferentially engages the RG conformation and produces a shorter cAMP response. Labelled doses differ (20 µg versus 80 µg daily) and the patent positions differ.
Is glycopyrrolate a peptide?
No. Glycopyrrolate (CAS 596-51-0, C19H28BrNO3, 398.33 g/mol) is a synthetic quaternary ammonium anticholinergic produced by esterification and quaternisation, purified by crystallisation and stored at ambient temperature. It is included in this comparison because it is frequently quoted alongside peptide APIs in the same enquiries, and because it establishes the contrast: it requires roughly 300 times more API mass per treated patient-year than teriparatide while carrying no cold chain and no compound patent.
What is the CAS number and molecular weight of glycopyrrolate?
Glycopyrronium bromide has CAS 596-51-0, molecular formula C19H28BrNO3 and molecular weight 398.33. The active moiety, the glycopyrronium cation, is 318.43 g/mol. The FDA label states that glycopyrronium constitutes 80% of the nominal tablet strength, so 1 mg of glycopyrronium bromide corresponds to 0.8 mg of glycopyrronium. Glycopyrrolate is the United States Adopted Name; glycopyrronium is the International Nonproprietary Name for the same substance.
What is the HS code for semaglutide and tirzepatide API?
United States Customs has ruled that both fall in HTSUS 2937.19.0000, duty free: semaglutide in bulk form under ruling NY N341149 (2024) and tirzepatide in bulk powder form under ruling N341897 (16 August 2024). The subheading covers polypeptide hormones, protein hormones and glycoprotein hormones, their derivatives and structural analogues, other than somatotropin and insulin. Finished dosage forms move to Chapter 30 instead.
What is the HS code for glycopyrrolate?
CBP ruling 886335 classified bulk glycopyrrolate (CAS 596-51-0) in HTSUS 2933.90.7500, heterocyclic compounds with nitrogen hetero-atom(s) only, other, aromatic or modified aromatic, drugs, at 6.9% ad valorem under that ruling. Glycopyrrolate in dosage form has been classified in Chapter 30, most recently under statistical suffix 3004.90.9257, established for products containing glycopyrrolate or its salts effective 1 February 2026. Classification is confirmed per destination at the time of entry.
When did the semaglutide patent expire in China?
The core compound patent expired in China on 20 March 2026. Protection in the United States, Japan and Europe runs to 2031–2032, and formulation patents in some jurisdictions extend as far as 2042. Following the China expiry, more than ten domestic manufacturers had filed generic or biosimilar applications, and chemically-modified generics were expected to receive approval from the third quarter of 2026. The originator has announced price reductions of 35–50% for 2027.
Why do peptide APIs cost so much more per kilogram than glycopyrrolate?
The difference is manufacturing technology. Glycopyrrolate is assembled in two bond-forming operations and purified by crystallisation. Semaglutide and tirzepatide require 30 to 38 successive coupling cycles on a solid support, followed by lipidation, preparative reversed-phase HPLC and lyophilisation. Yield compounds across the chain — 99.5% per step over 38 couplings returns roughly 83% before purification — and purification throughput, measured in column-hours, is the binding capacity constraint rather than reactor volume.
What is the difference between HPLC purity and peptide content?
HPLC purity describes the main-peak area as a percentage of total peak area in a chromatographic run. Peptide content is the mass fraction of the actual peptide in the material. Water and counter-ion (acetate or trifluoroacetate) remaining after lyophilisation routinely account for 10–20% of a lyophilised peptide's mass, so a material reporting HPLC purity above 99% can still have a peptide content near 80–90%. Both figures, together with water content by Karl Fischer and counter-ion identity, belong on the specification.
How should peptide API be stored and shipped?
Lyophilised peptide powders are held frozen at −20 °C, sealed, dry and protected from light; supplier stability statements for teriparatide powder reach four years or more at that temperature. Short ambient transit is generally tolerated by lyophilised powder. Finished injectable products are refrigerated at 2–8 °C, must not be frozen, and — for teriparatide — are capped after use because the solution is light-sensitive, with in-use stability demonstrated for 28 days. Repeated freeze–thaw of reconstituted solutions causes incremental inactivation, so aliquoting after first dissolution is standard.
Can semaglutide or tirzepatide API still be supplied to compounding pharmacies in the United States?
The shortage-driven enforcement discretion window has closed. The FDA removed tirzepatide injection from the shortage list in October 2024 and semaglutide injection in December 2024. Enforcement discretion then ended for 503A pharmacies compounding tirzepatide in February 2025, 503B outsourcing facilities compounding tirzepatide in March 2025, 503A pharmacies compounding semaglutide in April 2025 and 503B facilities compounding semaglutide in May 2025. API supplied into a compounding channel after those dates occupies a different regulatory position from API supplied to a licensed manufacturer holding an open DMF or CEP.
Which of the five has the largest export volume?
Semaglutide, by a wide margin, on a value-weighted basis. Tirzepatide follows with comparable demand but far tighter lawful supply because its compound patents run to 2036. Glycopyrrolate, teriparatide and abaloparatide follow at a much lower level. The ranking inverts partially if measured in shipped mass: glycopyrrolate requires roughly 2,190 mg of API per treated patient-year against 7.3 mg for teriparatide, so a kilogram-weighted index places the small molecule near the top and the bone peptides at the bottom.
How much API does one treated patient consume in a year?
From published label regimens, before titration waste, overfill and process yield: glycopyrrolate at 2 mg three times daily gives approximately 2,190 mg per year; tirzepatide at 15 mg weekly gives approximately 780 mg; semaglutide at 2.4 mg weekly gives approximately 125 mg; abaloparatide at 80 µg daily gives approximately 29.2 mg; teriparatide at 20 µg daily gives approximately 7.3 mg. The roughly 300-fold spread is the reason the incretins compete for tonne-scale purification capacity while the bone peptides are served by kilogram campaigns.

Request a Quote →17. Conclusion
Five molecules, and the most useful thing that can be said about them is that they do not belong to one market. Glycopyrrolate is a 398-dalton quaternary ammonium salt finished in two operations, shipped at room temperature, free of compound patent since the 1960s, and the heaviest API per treated patient-year in the group at roughly 2.19 grams. Teriparatide and abaloparatide are 34-residue PTH1R agonists dosed in micrograms, one post-patent since 2019 and one RG-selective, together requiring single-digit to tens of kilograms of API per year. Semaglutide and tirzepatide are lipidated incretin peptides dosed in milligrams, and their demand is what moved China's peptide sector from hundred-kilogram to tonne-scale manufacturing inside three years.
Three facts decide most sourcing outcomes in this group. Patent position is jurisdictional, not molecular — semaglutide's China compound patent lapsed in March 2026 while the United States compound patent runs to December 2031 and tirzepatide's runs to 2036 in both. Mass basis is explicit or it is ambiguous — glycopyrrolate has a bromide and a cation basis differing by 20%, and every peptide carries a counter-ion and a water content that separate chromatographic purity from actual peptide content. Capacity is a purification constraint, not a synthesis constraint — reactor litres and column-hours, and a 300-fold spread in API mass per treated patient-year between the top and bottom of the group.
Everything else — cold chain, documentation depth, lead time and price per gram — follows from which of those three facts applies.
